چکیده

Salinomycin is a carboxylic polyether ionophore which was isolated from the culture super‐ natant of the bacterium Streptomyces albus in 1974 [1]. Structurally, it is composed of a penta‐ cyclic molecule with a unique tricyclic spiroketal ring system and a unsaturated sixmembered ring (Fig. 1). Its lipophilic property enables salinomycin to act in cytoplasmic and mitochondrial membranes as an ionophore with a strong preference for potassium. There‐ fore, it promotes cellular and mitochondrial potassium efflux and inhibits mitochondrial ox‐ idative phosphorylation [2, 3].

Evaluating the Effects of Separate and Concomitant Use of MK-2206 and Salinomycin on Prostate Cancer Cell Line

Background: Prostate cancer is known as one of the most prevalent health disorders in the male population globally. The aim of the current study was to evaluate the effects of separate and concomitant use of MK-2206 and salinomycin on prostate cancer cell line.  Methods: The antitumor potential of separate and concomitant use of MK-2206 and salinomycin was evaluated in a panel of prostate ca...

Inhibition of Autophagy Promotes Salinomycin-Induced Apoptosis via Reactive Oxygen Species-Mediated PI3K/AKT/mTOR and ERK/p38 MAPK-Dependent Signaling in Human Prostate Cancer Cells

Recently, the interplay between autophagy and apoptosis has become an important factor in chemotherapy for cancer treatment. Inhibition of autophagy may be an effective strategy to improve the treatment of chemo-resistant cancer by consistent exposure to chemotherapeutic drugs. However, no reports have clearly elucidated the underlying mechanisms. Therefore, in this study, we assessed whether s...

Salinomycin Exerts Anticancer Effects on PC-3 Cells and PC-3-Derived Cancer Stem Cells In Vitro and In Vivo

Salinomycin is an antibiotic isolated from Streptomyces albus that selectively kills cancer stem cells (CSCs). However, the antitumor mechanism of salinomycin is unclear. This study investigated the chemotherapeutic efficacy of salinomycin in human prostate cancer PC-3 cells. We found that cytotoxicity of salinomycin to PC-3 cells was stronger than to nonmalignant prostate cell RWPE-1, and expo...

P-49: Effect of Crab Shell HydroalcoholicExtract Induced Apoptosis in ProstateCancer Cell Line

Background: Prostate cancer is the second most common cancer in men worldwide and the third most common cancer in Iranian men. Chemotherapy is used to treat this cancer, but it has toxic effect. Natural compounds found in marine organisms and plants have anticancer considered. The aim of the present study was to investigate the effect of crab shell extract on induction of apoptosis in prostate ...

Salinomycin induces cell death with autophagy through activation of endoplasmic reticulum stress in human cancer cells

Salinomycin is perhaps the first promising compound that was discovered through high throughput screening in cancer stem cells. This novel agent can selectively eliminate breast and other cancer stem cells, though the mechanism of action remains unclear. In this study, we found that salinomycin induced autophagy in human non-small cell lung cancer (NSCLC) cells. Furthermore, we demonstrated tha...
